The second domain is a Ca2+\/calmodulin-dependent protein kinase II (CaMKII)-specific inhibitor. CaMKII is a promising and extensively investigated target for neuroprotection after ischemic insult. Pharmacokinetic (PK) studies demonstrate a half-life of 10-15 min in vivo, which is well-suited for clinical use. TatCN19o is protected by four issued US patents.<\/p><p>Current proof-of-concept results include:<\/p><p><u>Rodent<\/u><\/p><ul><li>Neuroprotective after cardiac arrest and CPR<\/li><li>Neuroprotective after ischemic stroke<\/li><li>Works in combination with therapeutic hypothermia standard of care<\/li><li>Treatment prevented ischemia-induced loss of synaptic plasticity and protected memory function<\/li><\/ul><p><u>Pig<\/u><\/p><ul><li>Neuroprotective after ventricular fibrillation and return of spontaneous circulation<\/li><\/ul><p><u>Safety<\/u><\/p><ul><li>No acute adverse effects in mice at 1,000x therapeutic dose<\/li><li>No acute adverse effects in pigs at 20x therapeutic dose<\/li><li><em>In vitro<\/em> studies showed no off-target binding<\/li><li>No adverse effects on memory<\/li><\/ul><p>Together, these results demonstrate a safe and robust neuroprotective effect, even when the drug is administered hours after acute neuronal injury.
